虾青素对运动性力竭大鼠肾脏铁死亡的影响及机制研究

摘要:

探讨虾青素对运动性力竭大鼠肾脏铁死亡的影响及潜在调控机制,研究选取60只8周龄SD雄性大鼠,开展为期4周的力竭训练实验用于建立力竭性大鼠模型。除对照组外,将建模成功的大鼠随机分为模型组、25mg/kg虾青素组、50mg/kg虾青素组、100mg/kg虾青素组和ML385(Nrf2抑制剂)+虾青素(100mg/kg)组。4周后,收集血液和肾脏样品并检查相关指标,结果发现50和100 mg/kg虾青素处理后,运动性力竭大鼠血清中尿素氮(BUN)、酸酐(SCR)和肾损伤分子1(KIM-1)水平显著降低,肾脏病理损伤明显改善·肾组织中的活性氧(ROS)、Fe2+、8-OHdG和丙二醛(MDA)含量显著降低,谷胱甘肽(GSH)和超氧化物歧化酶(SOD)水平显著升高,核因子E2相关因子2(Nrf2)、血红素加氧酶-1(HO-1)、溶质载体家族7成员11(SLC7A11)和谷胱甘肽过氧化物酶4(GPX4)蛋白表达水平增加,Kelch样环氧氯丙烷相关蛋白1(Keapl1)表达水平降低。而ML385干预逆转了虾青素对运动性力竭大鼠肾脏铁死亡相关指标和Nrf2/HO-1信号通路的影响.研究表明虾青素可能通过激活Nr2/HO-1信号通路来抑制运动性力竭大鼠肾脏铁死亡。

To investigate the effects of astaxanthin on renal ferroptosis in exercise-induced exhausted rats and its potential regulatory mechanisms, sixty 8-week-old male Sprague Dawley(SD) rats were selected for a 4-week exhaustion training ex-periment to establish the exhaustion rat model. Except for the control group, the successfully modelled rats were randomly di-vided into a model group, 25mg/kg astaxanthin group, 50mg/kg astaxanthin group, 100mg/kg astaxanthin group, and ML385 (Nrf2 inhibitor) plus astaxanthin(100mg/kg) group. After 4 weeks, blood and kidney samples were collected to examine relevant indicators. The results showed that treatments with 50 and 100mg/kg astaxanthin significantly reduced serum bood urea nitrogen(BUN). creatinine(SCR), and kidney injury molecule 1(KIM-1) levels in exercise-induced exhausted rats.markedly improved kidney pathological damage, significantly decreased reactive oxygen species (ROS), Fe2t, 8-OHdG and MDA contents in renal tissue, and significantly increased glutathione(GSH) and superoxide dismutase(SOD) levels. Addition-ally, protein expression levels of nuclear factor E2-related factor 2(Nrf2), haem oxygenase-1(HO-1), solute carrier family 7 member 11(SLC7A11), and glutathione peroxidase 4(GPX4) increased, while Kelch-like ECH-associated protein 1(Keap1)expression decreased. ML385 intervention reversed the effects of astaxanthin on renal ferroptosis-related indicators and the Nrf2/HO-1 signalling pathway in exercise-induced exhausted rats. The study indicates that astaxanthin may inhibit renal fer-roptosis in exercise-induced exhausted rats by activating the Nrf2/HO-1 signalling pathway.
