The interaction of histone H2A.W and AGO2 in Arabidopsis thaliana

以拟南芥AGO2为研究对象,通过免疫共沉淀质谱联用技术(IP-MS),发现了HTA6和HTA7可能与AGO2蛋白互作,并通过亚细胞定位、双分子荧光互补技术以及免疫共沉淀(Co-IP)实验,进一步研究了H2A.W家族蛋白HTA6和HTA7与AGO2蛋白的定位和相互作用关系.结果显示HTA6,HTA7和 AGO2在细胞核内有共定位,且HTA6,HTA7分别与AGO2在细胞核内存在相互作用,体内Co-IP实验也进一步证明了HTA6.HTA7和AGO2的互作关系,为后续研究AGO2蛋白与异染色质之间的调控关系奠定了基础.
Taking Arabidopsis thaliana AGO2 as the research object, through the immunoprecipitation-mass spectrom-etry(IP-MS) technique, HTA6 and HTA7 were found to potentially interact with AGO2 protein. Further studies on the locali-zation and interaction relationship between H2A. W family protcins HTA6 and HTA7 and AGO2 protcin were conducted using subcellular localization, bimolecular fluorescence complementation technology and co-immunoprecipitation(Co-IP) experiments.The results showed that HTA6, HTA7 and AGO2 were co-localized in the nucleus, and HTA6 and HTA7 respectively inter-acted with AGO2 in the nucleus. The in vivo Co-IP experiment further confirmed the interaction relationship among HTA6.HTA7 and AGO2, laying a foundation for subsequent research on the regulatory relationship between AGO2 protein and heter ochromatin.
